Tesamorelin is a synthetic analog of growth hormone-releasing hormone (GHRH), modified at the N-terminus for resistance to enzymatic degradation. It's one of the most extensively characterized GHRH analogs in published clinical literature, with a well-established pharmacodynamic profile from pooled Phase 3 trial data.
Mechanism of Action
Tesamorelin binds the GHRH receptor on pituitary somatotrophs, stimulating pulsatile endogenous growth hormone release. Research has used it to characterize GHRH receptor signaling, downstream IGF-1 axis response, and the pharmacokinetic effect of N-terminal stabilization (a trans-3-hexenoic acid modification) on GHRH analog half-life relative to native GHRH(1-44).
