GLP-1 S is a long-acting GLP-1 receptor agonist and the most extensively characterized incretin peptide in published clinical literature, with over a decade of receptor-binding, pharmacokinetic, and metabolic signaling data behind it. It serves as the reference compound against which newer multi-receptor agonists are typically benchmarked in comparative research.
Mechanism of Action
GLP-1 S binds and activates the GLP-1 receptor, an Aib8 substitution conferring resistance to DPP-4 cleavage and a C18 fatty diacid chain (attached via an AEEA-AEEA-γGlu linker) extending its plasma half-life. Research has used it to characterize GLP-1 receptor binding kinetics, downstream cAMP signaling, and the pharmacokinetic profile of fatty-acid-conjugated peptide half-life extension as a platform technology.
